Clinical development of this compound followed earlier short-acting GLP-1 analogues that required frequent injection. Once-weekly subcutaneous formulations entered use after 2017, and an oral formulation using a permeation enhancer later became available. The oral version pairs the peptide with sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, a carrier that improves uptake across the gastric epithelium. Interest has expanded from glycaemic control into weight management and metabolic liver disease. Regulatory status and approved indications differ between countries, and the product remains subject to ongoing safety monitoring.
Semaglutide is a synthetic peptide that acts as an agonist at the glucagon-like peptide-1 receptor. It is a structural analogue of human GLP-1(7-37), modified to resist enzymatic degradation by dipeptidyl peptidase-4. The peptide backbone contains alpha-aminoisobutyric acid at position 8, a substitution that stabilises the helix and slows cleavage. A fatty diacid side chain attached through a linker at lysine 34 promotes binding to serum albumin, which extends the circulating half-life. These two modifications together allow less frequent administration than native GLP-1 requires.
Activation of the GLP-1 receptor couples to Gs signalling and raises intracellular cyclic AMP in pancreatic beta cells. The resulting insulin release depends on prevailing glucose concentrations, so the effect is greater when glucose is elevated. Receptor engagement also suppresses glucagon secretion and slows gastric emptying, which flattens post-meal glucose excursions. In the central nervous system, signalling in hypothalamic and brainstem regions is associated with reduced appetite and lower energy intake. Studies continue to examine effects on cardiac, renal and hepatic endpoints; whether those benefits are independent of weight change remains an open question.

Receptor activation raises intracellular cyclic AMP through Gs coupling, which promotes glucose-dependent insulin release and suppresses glucagon secretion when blood glucose is elevated. Effects outside the pancreas include slower gastric emptying and altered appetite signalling in the hypothalamus and hindbrain. The relative contribution of each tissue to overall metabolic outcomes remains an area of active investigation. Central mechanisms in particular are inferred mainly from animal models and indirect human measures rather than direct observation.
Serum protein binding dominates the pharmacokinetic profile. The attached chain associates strongly with albumin, shielding the peptide from enzymatic attack and slowing filtration by the kidney. This interaction extends the circulation half-life to roughly one week in humans, which supports weekly administration intervals. An oral version pairs the peptide with an absorption enhancer that transiently alters gastric epithelium, permitting limited uptake; bioavailability by that route is substantially lower than by injection.
Receptor binding triggers G protein signaling that raises intracellular cyclic AMP in pancreatic beta cells. Insulin release follows in a glucose-dependent manner, so secretion increases when blood glucose is elevated and diminishes when it is not. The same signaling suppresses glucagon release from alpha cells and slows gastric emptying, which blunts the post-meal glucose rise. In the brain, receptor activation in regions such as the arcuate nucleus is associated with reduced appetite and lower energy intake. How much each of these effects contributes to overall weight change is not fully settled.
Two structural features account for the prolonged half-life of semaglutide. A modified amino acid at position 8 resists cleavage by dipeptidyl peptidase-4, the enzyme that rapidly degrades native GLP-1. A fatty diacid side chain binds serum albumin, which limits renal clearance and protects the peptide from enzymatic breakdown. These modifications yield a plasma half-life of approximately one week in humans, allowing once-weekly administration. The relationship between plasma concentration and clinical effect varies between individuals, and sources of that variability are still being characterized.
Semaglutide is a synthetic peptide analog of glucagon-like peptide-1 (GLP-1), a hormone released from intestinal L-cells after food intake. The compound belongs to the incretin mimetic class and acts at GLP-1 receptors distributed across pancreatic, gastrointestinal, cardiovascular, and central nervous system tissues. Compared with native GLP-1, the molecule carries structural changes that extend its activity from minutes to roughly one week. It is studied for glycemic control in type 2 diabetes and for weight management, and its effects on cardiovascular and other outcomes remain active research areas.

Commonly served on a Sunday or any other special occasion, it can be done in a kettle BBQ or a conventional oven. Typical preparation involves covering the leg of lamb with butter, pushing rosemary sprigs into incisions cut in the leg, and sprinkling rosemary leaves on top. The lamb is then roasted for two hours at 180 °C (350 °F) and typically served with carrots and potato (also roasted), green vegetables and gravy. In Indonesia, lamb is popularly served as lamb satay and lamb curry. Both dishes are cooked with various spices from the islands, and served with either rice or lontong. A version of lamb and bamboo shoot curry is the specialty of Minang cuisine, although similar dishes can be found in Thai cuisine. In Mexico, lamb is the meat of choice for barbacoa, in which the lamb is roasted or steamed wrapped in maguey leaves underground. In Japan, although lamb is not traditionally consumed in most of the country, on the northern island of Hokkaido and in the northeastern Tohoku regions, a hot-pot dish called jingisukan (i.e. "Genghis Khan") is popular. In that dish, thin-sliced lamb is cooked over a convex skillet alongside various vegetables and mushrooms in front of the diners, then dipped in soy-sauce-based dipping sauces and eaten. It was so named because lamb is popular in Mongolia (see "Sheep meat consumption" above).
Plasma was already well known when described by William Harvey in de Motu Cordis in 1628, but knowledge of it probably dates as far back as Vesalius (1514–1564). The discovery of fibrinogen by William Henson, c. 1770, made it easier to study plasma, as ordinarily, upon coming in contact with a foreign surface – something other than the vascular endothelium – clotting factors become activated and clotting proceeds rapidly, trapping RBCs etc. in the plasma and preventing separation of plasma from the blood. Adding citrate and other anticoagulants is a relatively recent advance. Upon the formation of a clot, the remaining clear fluid (if any) is blood serum, which is essentially plasma without the clotting factors The use of blood plasma as a substitute for whole blood and for transfusion purposes was proposed in March 1918, in the correspondence columns of the British Medical Journal, by Gordon R. Ward. "Dried plasmas" in powder or strips of material format were developed and first used in World War II. Prior to the United States' involvement in the war, liquid plasma and whole blood were used.
== Causes == Dominant genetic disorders can be caused by just a single copy of an abnormal gene. This abnormal gene can be the result of being inherited from either parent or be a new mutation. Most cases are caused by a de novo (new) mutation in the gene that occurs during the formation of the egg or sperm. These cases occur when there is no history of the disorder in the family. The COL11A2 gene is responsible for providing instructions on making one component of the type XI collagen. Type XI collagen is a complex molecule that helps give structure and strength to the connective tissues. Collagen is found in bone. It is also found in cartilage that makes up most of the skeleton during early development. The mutation of COL11A2 in Weissenbacher-Zweymüller syndrome disrupts the assembly of the type XI collagen molecules. The malfunctioning collagen weakens the connective tissue causing impaired bone development. COL11A2 is also associated with autosomal dominant non-syndromic hearing loss (ADNSHL). All mutations of COL11A2 in ADNSHL are missense mutations.
Technological utopianism refers to the belief that technological development is a moral good, which can and should bring about a utopia, that is, a society in which laws, governments, and social conditions serve the needs of all its citizens. Examples of techno-utopian goals include post-scarcity economics, life extension, mind uploading, cryonics, and the creation of artificial superintelligence. Major techno-utopian movements include transhumanism and singularitarianism. The transhumanism movement is founded upon the "continued evolution of human life beyond its current human form" through science and technology, informed by "life-promoting principles and values". The movement gained wider popularity in the early 21st century. Singularitarians believe that machine superintelligence will "accelerate technological progress" by orders of magnitude and "create even more intelligent entities ever faster", which may lead to a pace of societal and technological change that is "incomprehensible" to humanity. This event horizon is known as the technological singularity. Major figures of techno-utopianism include Ray Kurzweil and Nick Bostrom. Techno-utopianism has attracted both praise and criticism from progressive, religious, and conservative thinkers.

The Maronite Catholic and the Druze founded modern Lebanon in the early eighteenth century, through the ruling and social system known as the "Maronite-Druze dualism" in Mount Lebanon Mutasarrifate. Contact between Christians (members of the Maronite, Eastern Orthodox, Melkite, and other churches) and the Unitarian Druze led to the presence of mixed villages and towns in Mount Lebanon, Chouf, Wadi al-Taym, Jabal al-Druze, the Galilee region, Mount Carmel, and Golan Heights. They both speak the Arabic language and follow a social patterns similar to those of other peoples in the Levant (Eastern Mediterranean). Scholars classify Antiochian Greek Christians, Druze, and Maronites as ethnoreligious groups. The relationship between the Druze and Christians in Syria, Lebanon and Israel has been characterized by harmony and peaceful coexistence. Historically, they lived in the Chuf Mountains in complete harmony. On a social level, Druze and Christians have often interacted in daily life, sharing neighborhoods, schools, and workplaces. Druze and Christians in these regions celebrate each other's births, weddings, funerals, and celebrations such as Christmas, Maundy Thursday (in Lebanon), Easter and the Christian festival of Saint Elias (in Mount Carmel). Before 2011, more than 55,000 Christians lived in As-Suwayda Governorate, the only governorate in Syria with a Druze majority. In 2010, more than 52,000 registered Christian voters, primarily Maronites and Greek Orthodox, lived in the Aley District, where Druze constitute the majority.
== Data management == The FDA has collected a large amount of data through the decades. The OpenFDA project was created to enable easy access of the data for the public and was officially launched in June 2014.
A mass spectrum is a histogram plot of intensity vs. mass-to-charge ratio (m/z) in a chemical sample, usually acquired using an instrument called a mass spectrometer. Not all mass spectra of a given substance are the same; for example, some mass spectrometers break the analyte molecules into fragments; others observe the intact molecular masses with little fragmentation. A mass spectrum can represent many different types of information based on the type of mass spectrometer and the specific experiment applied. Common fragmentation processes for organic molecules are the McLafferty rearrangement and alpha cleavage. Straight chain alkanes and alkyl groups produce a typical series of peaks: 29 (CH3CH2+), 43 (CH3CH2CH2+), 57 (CH3CH2CH2CH2+), 71 (CH3CH2CH2CH2CH2+) etc.
Pizotifen, also known as pizotyline and sold under the brand names Sandomigran and Mosegor among others, is an antimigraine agent of the tricyclic group which is used primarily as a preventative to reduce the frequency of recurrent migraine headaches.
== Prognosis == Individuals with BED often have a lower overall quality of life and face social difficulties. Early behavior changes can predict a full recovery for the future. Individuals who have BED commonly have other conditions such as depression, personality disorder, bipolar disorder, substance abuse, body dysmorphic disorder, kleptomania, irritable bowel syndrome, fibromyalgia, or an anxiety disorder. They may also have history of attempted suicide and reoccurring panic attacks. While people of a normal weight may overeat occasionally, an ongoing habit of consuming large amounts of food in a short period of time may ultimately lead to weight gain and obesity. The main physical health consequences of this type of eating disorder are brought on by the weight gain resulting from calorie-laden bingeing episodes. Mental and emotional consequences of binge eating disorder include social weight stigma and emotional loss of control. Up to 70% of individuals with BED may also be obese, and therefore obesity-associated morbidities such as high blood pressure and coronary artery disease, type 2 diabetes mellitus, gastrointestinal issues (e.g., gallbladder disease), high cholesterol levels, musculoskeletal problems and obstructive sleep apnea may also be present. One study found a 42% obesity rate in those who have received a BED diagnosis. Additionally, a higher morbid obesity prevalence was observed in this population compared to a population without eating disorders.

Treatment may include dietary changes, iron supplements, and dealing with underlying causes, for example, medical treatment for parasites or surgery for ulcers. Supplementation with vitamin C may be recommended due to its potential to aid iron absorption. Severe cases may be treated with blood transfusions or iron infusions. Iron-deficiency anemia affected about 1.48 billion people in 2015. A lack of dietary iron is estimated to cause approximately half of all anemia cases globally. Women and young children are most commonly affected. In 2015, anemia due to iron deficiency resulted in about 54,000 deaths – down from 213,000 deaths in 1990.
The UK government announces it will overhaul what it describes as the "feudal" leasehold system in England and Wales by the end of the current parliament. A security incident at Buchanan bus station in the centre of Glasgow leads to the evacuation of the area and anti-terror police being called out; around three men are arrested in connection with the incident. 22 November Ofgem announces the price of an average energy bill will rise by £21 a year from January 2025, with the typical household paying £1,738 per year; prices are now 50% higher than pre-COVID levels. Data from the Office for National Statistics shows a 0.7% fall in retail sales during October, with people holding back on spending ahead of the budget. Downing Street indicates that Israeli Prime Minister Benjamin Netanyahu would face arrest if he travelled to the UK following the issuing of an international arrest warrant for war crimes by the International Criminal Court. The Investigatory Powers Tribunal rules that 300 people affected by the Manchester Arena bombing cannot continue with a case against MI5 in which they alleged their human rights were breached because the security agency failed to take "appropriate measures" to prevent the attack. The UK government announces plans for new "respect orders" in England and Wales, giving police and local authorities the power to ban those who persistently practice antisocial behaviour from drinking in public places or from entering town centres, with prison sentences of up to two years for those who consistently break the orders.
== Structure == The rectus abdominis is a very long flat muscle, which extends along the whole length of the front of the abdomen, and is separated from its fellow of the opposite side by the linea alba. Tendinous intersections (intersectiones tendineae) further subdivide each rectus abdominis muscle into a series of smaller muscle bellies. Tensing of the rectus abdominis causes the muscle to expand between each tendinous intersection. The upper portion, attached principally to the cartilage of the fifth rib, usually has some fibers of insertion into the anterior extremity of the rib itself.
== Mechanism == The creatine phosphate system is needed for the storage and transmission of phosphate-bound energy in the brain and muscle. The brain and muscle have particularly high metabolic demands, therefore, making creatine a necessary molecule in ATP homeostasis. In regard to the brain, in order for creatine to reach the brain, it must first pass through the blood–brain barrier (BBB). The BBB separates blood from brain interstitial fluid and is, therefore, able to regulate the transfer of nutrients to the brain from the blood. In order to pass through the BBB, creatine utilizes creatine transporter (CRT). When present at the BBB, CRT mediates the passage of creatine from the blood to the brain. When being transported from the blood to the brain, creatine has to constantly move against the creatine concentration gradient that is present at the border between the brain and circulating blood.

It belongs to the incretin mimetic class and acts as a long-acting glucagon-like peptide-1 receptor agonist. The class includes several peptides with different half-lives and routes of administration.
