The POI is isolated, typically by SDS-PAGE or chromatography. The isolated POI may be chemically modified to stabilise Cysteine residues (e.g. S-amidomethylation or S-carboxymethylation). The POI is digested with a specific protease to generate peptides. Trypsin, which cleaves selectively on the C-terminal side of Lysine or Arginine residues, is the most commonly used protease. Its advantages include i) the frequency of Lys and Arg residues in proteins, ii) the high specificity of the enzyme, iii) the stability of the enzyme and iv) the suitability of tryptic peptides for mass spectrometry. The peptides may be desalted to remove ionizable contaminants and subjected to MALDI-TOF mass spectrometry. Direct measurement of the masses of the peptides may provide sufficient information to identify the protein (see Peptide mass fingerprinting) but further fragmentation of the peptides inside the mass spectrometer is often used to gain information about the peptides’ sequences. Alternatively, peptides may be desalted and separated by reversed phase HPLC and introduced into a mass spectrometer via an ESI source. LC-ESI-MS may provide more information than MALDI-MS for protein identification but uses more instrument time. Depending on the type of mass spectrometer, fragmentation of peptide ions may occur via a variety of mechanisms such as collision-induced dissociation (CID) or post-source decay (PSD). In each case, the pattern of fragment ions of a peptide provides information about its sequence.
Constancy testing is the verification of reference values as part of quality assurance in x-ray diagnostics, nuclear medicine diagnostics, and radiotherapy. National regulations specify which parameters are to be tested, which limits are to be observed, which test methods are to be used, and which test samples are to be used. In Germany, the Radiation Protection in Medicine Directive and the relevant DIN 6855 standard in nuclear medicine require regular (in some cases daily) constancy testing. Test sources are used to check the response of probe measuring stations as well as in vivo and in vitro measuring stations. All-cause mortality is higher above 64 mmol/mol (8.0 DCCT%) HbA1c as well as below 42 mmol/mol (6.0 DCCT %) in diabetic patients, and above 42 mmol/mol (6.0 DCCT %) as well as below 31 mmol/mol (5.0 DCCT %) in non-diabetic persons, indicating the risks of hyperglycemia and hypoglycemia, respectively. Similar risk results are seen for cardiovascular disease. The 2022 ADA guidelines reaffirmed the recommendation that HbA1c should be maintained below 7.0% for most patients. Higher target values are appropriate for children and adolescents, patients with extensive co-morbid illness and those with a history of severe hypoglycemia. Solid-liquid extraction based ambient ionization uses a charged spray to create a liquid film on the sample surface. Molecules on the surface are extracted into the solvent. The action of the primary droplets hitting the surface produces secondary droplets that are the source of ions for the mass spectrometer. Desorption electrospray ionization (DESI) creates charged droplets that are directed at a solid sample a few millimeters to a few centimeters away. The charged droplets pick up the sample through interaction with the surface and then form highly charged ions that can be sampled into a mass spectrometer. Plasma-based ambient ionization is based on an electrical discharge in a flowing gas that produces metastable atoms and molecules and reactive ions. Heat is often used to assist in the desorption of volatile species from the sample. Ions are formed by chemical ionization in the gas phase. A direct analysis in real time (DART) source operates by exposing the sample to a dry gas stream (typically helium or nitrogen) that contains long-lived electronically or vibronically excited neutral atoms or molecules (or "metastables"). Excited states are typically formed in the DART source by creating a glow discharge in a chamber through which the gas flows.

Agriculture was once the most important industry in Denmark. Nowadays, it is of minor economic importance. In 2016, 62,000 people, or 2.5% of all employed people worked in agriculture and horticulture. Another 2,000 people worked in fishing. As value added per person is relatively low, the share of national value added is somewhat lower. Total gross value added in agriculture, forestry and fishing amounted to 1.6% of total output in Denmark (in 2017). Despite this, Denmark is still home to various types of agricultural production. Within animal husbandry, it includes dairy and beef cattle, pigs, poultry and fur animals – all sectors that produce mainly for export. Regarding vegetable production, Denmark is a leading producer of grass-, clover- and horticultural seeds. The agriculture and food sector as a whole represented 25% of total Danish commodity exports in 2015. 63% of the land area of Denmark is used for agricultural production – the highest share in the world according to a report from University of Copenhagen in 2017. The Danish agricultural industry is historically characterized by freehold and family ownership, but due to structural development farms have become fewer and larger. In 2020 the number of farms was approximately 33,000, of which approximately 10,000 were owned by full-time farmers.
== Further development == Partition chromatography allowed further developments of column chromatography, and inspired new forms of chromatography such as countercurrent distribution, paper chromatography, and gas chromatography. Commercial counter-current distribution instruments were used for many important discoveries The introduction of paper chromatography was an important analytical technique which gave rise to thin-layer chromatography. Finally, gas-liquid chromatography, a fundamental technique in modern analytical chemistry, was described by Martin with coauthors A. T. James and G. Howard Smith in 1952. The stationary phase can be changed to change the separation characteristics. By chemically bonding alkane functional groups to silica gel, we obtain reversed-phase chromatography. In 1944, Lyman C. Craig accomplished the original goal of Martin and Synge: column chromatography with two free-flowing liquid phases.
=== The Taghi organisation === Ridouan Taghi is a former Dutch-Moroccan drug trafficker and crime boss. He was specialised in forming criminal groups and coalitions that operated as international cocaine cartels. He is suspected of being behind the orchestration of several assassinations, attacks, and kidnappings in connection to the Mocro-War. Documents produced by the Drug Enforcement Administration (DEA) of the United States, and sent to the Dutch police, exposed what appeared to be a super drug cartel headed by Ridouan Taghi, alongside Raffaele Imperiale (Camorra's drugs and arms dealer), Daniel Kinahan (Irish reputed gang boss) and Edin Gačanin (Bosnian drug trafficker). The DEA estimated that around a third of all cocaine trafficking in Europe was done through his organisation, with a virtually monopoly on Peruvian cocaine. After the decryption of the telecommunication servers of Ennetcom by the Dutch and Canadian authorities, it also exposed a very close and personal friendship between Ridouan Taghi, and Chilean cartel boss Rico "El Rico" Eduardo Riquelme Vega, who was arrested in Santiago, Chile in 2017. Following the arrest of Benaouf A. in 2013, and the assassinations of two rival drug lords, Gwenette Martha and Samir Bouyakhrichan in 2014, Taghi was able to establish himself as one of the most powerful drug traffickers in Europe. The Dutch government also considered him to be "one of the most dangerous drug traffickers in Europe". Ridouan Taghi was able to keep a very low profile and run his operation unknowingly for many years.

== DNA analysis == The bone fragment, identified by the code DC1227 (GenBank Accession = KU131206) or as Denisova 11, may have originated from a human arm or leg. Prior to the extraction of material for genetic analysis, DC1227 weighed 1.68 g (0.059 oz), and had maximum dimensions of 24.7 mm (0.97 in) by 8.39 mm (0.330 in). In 2016, a team from the Max Planck Institute for Evolutionary Anthropology in Leipzig, Germany, led by paleogeneticists Viviane Slon and Svante Pääbo, extracted six DNA samples and produced ten libraries of complete genome sequences to an average coverage of 2.6-fold. Isolated mitochondrial DNA and nuclear DNA indicated the archaic human was of nearly equal proportion Neanderthal and Denisovan. A comparison of the specimen's X chromosomes and autosomes showed it belonged to a female. The estimated heterozygosity of Denisova 11, found to be comparable to present day Africans suggests that the girl was a first-generation Neanderthal-Denisovan hybrid. Subsequent analyses showed a high likelihood that her Denisovan father also had some Neanderthal ancestry introduced into his genome hundreds of generations before his lifetime. Denisova 11's genome thus constitutes the first direct evidence for at least two instances of interbreeding between Neanderthals and Denisovans.
