fatty diacid raises a handful of sensible questions. This page answers them in order, starting with the fundamentals and moving to applications.

Receptor activation occurs at GLP-1 receptors distributed across pancreatic islets, the hypothalamus, and the gastrointestinal tract. Binding triggers G protein signaling that raises cyclic AMP and enhances glucose-dependent insulin release. Because the effect depends on prevailing glucose levels, insulin secretion does not rise when blood sugar is already low. Signaling in the brain and gut also influences appetite and gastric emptying, which is why the compound appears in both metabolic and weight-related research literature.
Development began in the early 2010s with the goal of extending GLP-1 activity beyond the brief window achieved by native peptide infusion. The earliest approved formulation was a subcutaneous injection given once weekly. A later oral tablet pairs the peptide with an absorption enhancer, sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, usually shortened to SNAC. That carrier lowers local pH and helps the peptide cross gastric tissue. Both routes deliver the same active molecule.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, a gut hormone released after meals. Its backbone retains the GLP-1 sequence but incorporates two substitutions that slow enzymatic breakdown by dipeptidyl peptidase-4. A short polyethylene glycol linker and a C18 fatty diacid are attached to the peptide chain, allowing the molecule to bind serum albumin and remain in circulation far longer than the native hormone. The result is a circulating half-life measured in days rather than the minutes typical of endogenous GLP-1.
The company that developed the compound filed it as a long-acting analogue, and it gained first approval in 2017 for type 2 diabetes. Later authorisations from several regulators extended the indication to chronic weight management, and the World Health Organization added the glucagon-like peptide-1 receptor agonist drug class to its model list of essential medicines in 2023. Production uses solid-phase peptide synthesis followed by side-chain conjugation and chromatographic purification. Supply constraints and cost differences across regions are well documented. Literature on long-term outcomes continues to grow, with many trials reporting surrogate endpoints rather than hard clinical endpoints.
Semaglutide is a synthetic peptide of thirty-one amino acids that shares roughly ninety-four percent sequence identity with human glucagon-like peptide-1. Two substitutions resist enzymatic cleavage by dipeptidyl peptidase-4, and a fatty diacid side chain attached through a linker promotes binding to serum albumin. That albumin binding slows renal clearance and extends the circulating half-life from minutes to approximately one week. The structural changes are well established in the published literature. Whether the same modifications affect receptor signalling bias in ways that matter clinically remains an open question.

Reversed-phase high-performance liquid chromatography is widely used to assess purity and to separate the parent peptide from related substances. Mass spectrometry confirms identity and can resolve modifications that differ by a few daltons. Size-exclusion chromatography detects dimers and higher aggregates, which are relevant to both stability and immunogenicity questions. Peptide mapping with enzymatic digestion locates specific modifications along the sequence. Circular dichroism provides a secondary-structure profile, although it gives limited information about local conformational changes.
Quality control for peptide material focuses on identity, purity, content and the profile of impurities. Common degradants include deamidated and oxidised forms, plus aggregates formed during storage or handling. Forced degradation studies under heat, light, acid and peroxide help define which conditions accelerate change and which analytical methods detect it. Limits for individual impurities are set by pharmacopoeial monographs or manufacturer specifications. How much a given impurity affects biological activity is often uncertain, and conclusions may depend on the assay used.
Stability studies examine how temperature, pH, and moisture influence degradation rates over time. In aqueous solution, hydrolysis and deamidation accelerate as pH moves away from mildly acidic conditions. Light exposure and residual metal ions can also trigger oxidation of susceptible residues. Accelerated aging at elevated temperature is used to estimate shelf life, though extrapolation to room temperature carries uncertainty because individual degradation pathways do not always scale predictably.
Lyophilized semaglutide powder is typically held at minus twenty degrees Celsius for long-term storage. At that temperature, solid-state degradation reactions proceed slowly and the peptide remains intact for extended periods. Repeated freeze-thaw cycles are best avoided because they promote aggregation and can shift the proportion of monomeric peptide present. Working aliquots are often prepared so that each portion is thawed only once, and desiccant is placed inside the vial to limit moisture uptake.
Handling guidance for research quantities calls for single-use aliquots, an inert atmosphere where practical, and avoidance of repeated freeze-thaw cycles that accelerate aggregation. Certificates of analysis typically report purity by peak area, water content, counter-ion identity, and residual solvent levels. In the scientific literature the compound is usually described by its full amino acid sequence, its registry number, or its structural class rather than by any proprietary label. Reporting standards vary between journals, and reviewers increasingly request raw chromatograms alongside tabulated purity figures. Whether current purity thresholds are adequate for every experimental context is debated.
Reversed-phase high-performance liquid chromatography with ultraviolet detection is the dominant approach for peptide purity assessment, usually paired with mass spectrometry to confirm molecular mass and sequence. Peptide mapping by enzymatic digestion and tandem mass spectrometry locates modifications such as deamidation and oxidation. Quantitation in plasma matrices can be performed by LC-MS/MS after solid-phase extraction. Method validation follows general guidance on accuracy, precision, linearity, and limits of detection. Comparability of results between laboratories, when no shared reference standard is available, remains an open question.
Semaglutide belongs to the glucagon-like peptide-1 receptor agonist class, a group of synthetic peptides that imitate an incretin hormone released by intestinal L cells after food intake. Native GLP-1 circulates for only a few minutes because dipeptidyl peptidase-4 cleaves it rapidly. The hormone acts on pancreatic islets, the gastrointestinal tract, and several brain regions. Because the natural peptide is short-lived, development work concentrated on analogues that keep receptor activity while resisting enzymatic breakdown and renal clearance.
The semaglutide sequence is a 31-residue analogue of human GLP-1, altered at three positions relative to the parent hormone. Aminoisobutyric acid replaces alanine at position 8, arginine replaces lysine at position 34, and a lipophilic diacid is attached to lysine 26 through a short linker. These features are reported consistently in the structural literature. The position 8 substitution blocks recognition by dipeptidyl peptidase-4, while the attached chain drives strong, reversible association with a carrier protein in blood.
Receptor activation raises intracellular cyclic AMP through Gs coupling, which promotes glucose-dependent insulin release and suppresses glucagon secretion when blood glucose is elevated. Effects outside the pancreas include slower gastric emptying and altered appetite signalling in the hypothalamus and hindbrain. The relative contribution of each tissue to overall metabolic outcomes remains an area of active investigation. Central mechanisms in particular are inferred mainly from animal models and indirect human measures rather than direct observation.
==== Multiple sclerosis-related fatigue ==== Fatigue is a common and often debilitating symptom experienced by people with multiple sclerosis. Reviews and meta-analyses of controlled trials have found that modafinil has modest effectiveness in managing MS-related fatigue, though improvements in fatigue severity scores have not consistently reached statistical significance. Optimal dosing and treatment schedules are not well established. Clinical assessments have found that adverse events were common. Most MS organizations are neutral on the off-label use of modafinil to alleviate fatigue associated with MS.
In the second case, a biosynthetic pathway needs to be engineered, for example, an E. coli strain that biosynthesizes a novel amino acid (p-aminophenylalanine) from basic carbon sources and includes it in its genetic code. Another example is the production of phosphoserine, a natural metabolite, which required alteration of its pathway flux to increase its production.
==== Point contact on a (3D) half-space ==== Analogously to the Flamant solution for the 2D half-plane, fundamental solutions are known for the linearly elastic 3D half-space as well. These were found by Boussinesq for a concentrated normal load and by Cerruti for a tangential load. See the section on this in Linear elasticity.

In the United States, the term "nickel" or "nick" originally applied to the copper-nickel Flying Eagle cent, which replaced copper with 12% nickel 1857–58, then the Indian Head cent of the same alloy from 1859 to 1864. Still later, in 1865, the term designated the three-cent nickel, with nickel increased to 25%. In 1866, the five-cent shield nickel (25% nickel, 75% copper) appropriated the designation, which has been used ever since for the subsequent 5-cent pieces. This alloy proportion is not ferromagnetic. The US nickel coin contains 0.04 ounces (1.1 g) of nickel, which at the April 2007 price was worth 6.5 cents, along with 3.75 grams of copper worth about 3 cents, with a total metal value of more than 9 cents. Since the face value of a nickel is 5 cents, this made it an attractive target for melting by people wanting to sell the metals at a profit. The United States Mint, anticipating this practice, implemented new interim rules on December 14, 2006, subject to public comment for 30 days, which criminalized the melting and export of cents and nickels. Violators can be punished with a fine of up to $10,000 and/or a maximum of five years in prison. As of February 19, 2025, the melt value of a US nickel (copper and nickel included) is $0.054 (108% of the face value).

=== Venoms === Certain types of venom, such as those produced by venomous snakes, can also cause proteolysis. These venoms are, in fact, complex digestive fluids that begin their work outside of the body. Proteolytic venoms cause a wide range of toxic effects, including effects that are:
=== 116th Congress (2019–2021) === In March 2019, Kennedy introduced the Holding Foreign Companies Accountable Act, which Trump signed into law on December 18, 2020. The law prohibits any company from listing on an American stock exchange if it refuses to allow the Public Company Accounting Oversight Board to audit its annual private audit for three consecutive years. It also requires companies to disclose whether they are owned by a foreign government. In 2019, Kennedy again introduced a bill to extend the National Flood Insurance Program. It extended the program through June 14, 2019, rather than lapsing on May 31. The bill passed the Senate on May 23, 2019, and was signed into law.
== External operations == The Selous Scouts were involved in the Rhodesian military's attacks on insurgents and their bases in neighbouring countries, often known as external operations (or 'externals' for short). These operations became frequent from 1976. The unit's role in external operations included intelligence collection and directly attacking insurgents. In the intelligence gathering role, the Selous Scouts' Reconnaissance Troop conducted one or two man long-range reconnaissance patrols to locate or gather information on insurgent bases in Mozambique and Zambia. During direct attacks the Selous Scouts often impersonated soldiers of the country they were operating in. The Selous Scouts operated in eastern Botswana. This included fighting small actions against insurgents and a raid in which ZAPU leaders were captured in Francistown. The Selous Scouts also used pseudo tactics to collect intelligence in Francistown. Operation Long John was launched on 25 June 1976, against two guerilla bases located in Mozambique. This operation used 'flying column' tactics for the first time, which involved six vehicles manned by the Selous Scouts attacking the bases. Large quantities of munitions were destroyed, but few casualties were inflicted. On 9 August 1976, the Selous Scouts carried out Operation Eland, a raid on a ZANLA and FRELIMO controlled refugee camp at Nyadzonia in Mozambique. The Selous Scouts, who were mostly black and disguised in FRELIMO uniforms, included former Portuguese Army soldiers and a former ZANLA commander.
Most cephalopods rely on vision to detect predators and prey and to communicate with one another. Consequently, cephalopod vision is acute: training experiments have shown that the common octopus can distinguish the brightness, size, shape, and horizontal or vertical orientation of objects. The morphological construction gives cephalopod eyes the same performance as shark eyes; however, their construction differs, as cephalopods lack a cornea and have an everted retina. Cephalopods' eyes are also sensitive to the plane of polarization of light. Unlike many other cephalopods, nautiluses do not have good vision; their eye structure is highly developed, but lacks a solid lens. They have a simple "pinhole" eye through which water can pass. Instead of vision, the animal is thought to use olfaction as the primary sense for foraging, as well as locating or identifying potential mates.

== Advantages == The SECT graft is a sort of hybrid procedure that combines the pedicle flap with the free gingival graft and enjoys the benefits of both. Pedicle flaps alone, such as the coronally advanced flap, frequently suffer from retraction and muscle pull.
An electron-dense granule defined by the presence of Toll-like receptor 9 (TLR9) and protein disulfide isomerase (PDI), termed the T granule, has also been described, although its existence remains controversial. PDI and other platelet-borne thiol isomerases have been reported to be packaged within a non-granular compartment derived from the megakaryocyte endoplasmic reticulum (ER), which may be associated with the dense tubular system.
The first Volunteer unit to be sent out was a 1,300 man composite battalion group, composed of infantry recruited from across London units and a field battery from the Honourable Artillery Company, the City Imperial Volunteers, which was raised in early January 1900; it was sent into combat after six weeks of training in South Africa, where Lord Roberts described it as "quite excellent", and was returned home in October. At the same time, a number of service companies were raised from volunteer units, employed as integral companies of their sister regular battalions, and were well regarded in the field. The decision was taken in late December to form a new force, the Imperial Yeomanry, to consist of mounted infantry. Whilst the Yeomanry provided many of the officers and NCOs, only a small number of the junior ranks came from existing Yeomanry regiments, with some more from Volunteer corps. The units performed well, but recruiting proceeded in fits and starts—recruitment stopped in May, and was only resumed in early 1901—and so an adequate supply of manpower was not always available. Sixty militia battalions, around 46,000 men, also volunteered and were eventually sent to South Africa. They were employed mainly on lines of communication, and regarded as second-line troops of low quality; this was unsurprising, as they were strongly deficient in officers, heavily composed of men of 18 and 19, who were regarded as too young by the Regular Army, with many of their best and most experienced men already deployed with regular units as members of the Militia Reserve.
=== CPT-II deficiency === Carnitine palmitoyltransferase II deficiency (also known as CPT-II deficiency) leads to an excess long chain fatty acids, as the body lacks the ability to transport fatty acids into the mitochondria to be processed as a fuel source. The disease is caused by a defect in the gene CPT2. This deficiency will present in patients in one of three ways: lethal neonatal, severe infantile hepatocardiomuscular, and myopathic form. The myopathic is the least severe form of the deficiency and can manifest at any point in the lifespan of the patient. The other two forms appear in infancy. Common symptoms of the lethal neonatal form and the severe infantile forms are liver failure, heart problems, seizures and death. The myopathic form is characterized by muscle pain and weakness following vigorous exercise. Treatment generally includes dietary modifications and carnitine supplements.
In Figure 5, both ribosomal subunits (small and large) assemble at the start codon (towards the 5' end of the mRNA). The ribosome uses tRNA that matches the current codon (triplet) on the mRNA to append an amino acid to the polypeptide chain. This is done for each triplet on the mRNA, while the ribosome moves towards the 3' end of the mRNA. Usually in bacterial cells, several ribosomes are working parallel on a single mRNA, forming what is called a polyribosome or polysome.

Native GLP-1 is degraded within minutes by circulating enzymes. The synthetic version carries substitutions at positions that block enzymatic cleavage, plus a fatty acid side chain that promotes albumin binding. These two changes together extend circulation time from minutes to roughly a week.
Albumin binding keeps a large fraction of the compound in a slowly released reservoir within the bloodstream. Plasma levels decline gradually rather than falling sharply after each administration. That profile supports dosing intervals measured in days instead of hours.
The active peptide sequence is the same in both formats. The oral version adds an absorption enhancer that is not present in the injected solution. Differences in excipients and formulation affect uptake rather than the identity of the active molecule.
It is a glucagon-like peptide-1 receptor agonist, often grouped with the incretin mimetics. Its backbone is modified from the human hormone to resist enzymatic degradation and to bind albumin. These two features distinguish it from the native peptide.
