Lyophilized semaglutide is typically stored at temperatures between minus 20 and minus 80 degrees Celsius for long-term preservation. Short-term storage at 2 to 8 degrees Celsius is common for working aliquots. Repeated freeze-thaw cycles can degrade the peptide and are usually avoided. The molecule is hygroscopic in its solid form, so containers should remain sealed with desiccant. Solutions are less stable than powders and are generally prepared fresh. Light exposure is limited because aromatic residues can undergo photo-oxidation.
Semaglutide dissolves readily in water and in aqueous buffers near neutral pH. Solubility decreases near the isoelectric point, where net charge is minimal. Common laboratory solvents include phosphate-buffered saline and dilute ammonium bicarbonate. Strongly acidic or basic conditions may accelerate hydrolysis. Working concentrations are usually prepared by diluting a concentrated stock. Vial surfaces can adsorb small amounts of peptide at low concentrations, so carrier proteins or low-binding tubes are sometimes used.
Reverse-phase high-performance liquid chromatography is the standard method for purity assessment, separating the peptide from truncated or oxidized variants. Mass spectrometry confirms molecular mass and detects modifications, while ultraviolet absorbance near 280 nanometers supports concentration measurement through tryptophan and tyrosine residues. Circular dichroism can indicate secondary structure, though the peptide is largely helical in solution, and ion-exchange chromatography resolves charge variants. Purity values above 95 percent are typical for research-grade material. Stability studies track degradation over time under defined conditions.
GLP-1 receptors are expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises cyclic AMP, enhances glucose-dependent insulin secretion, and suppresses glucagon release when blood glucose is high. Effects on gastric emptying and on hypothalamic appetite circuits reduce energy intake. Because insulin release remains glucose-dependent, the risk of hypoglycemia is low when the drug is used alone. The precise contribution of each pathway to body weight change in humans remains an area of active investigation.
Clinical studies of semaglutide generally measure glycated hemoglobin, fasting plasma glucose, body weight, and composite cardiovascular endpoints. The SUSTAIN program enrolled adults with type 2 diabetes, while the STEP program focused on obesity without diabetes. Administration follows a stepwise escalation schedule designed to limit gastrointestinal effects during the first weeks. Reported outcomes include mean percentage weight change, the proportion of participants reaching defined weight-loss thresholds, and rates of nausea, vomiting, and diarrhea. Long-term data on durability after treatment stops are still limited and remain a topic of ongoing research.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, developed by Novo Nordisk and first approved in 2017 for type 2 diabetes. It belongs to the incretin mimetic class, a group of agents that reproduce the glucose-dependent actions of endogenous GLP-1. The molecule was engineered to resist degradation by dipeptidyl peptidase-4 and to bind serum albumin, extending its half-life from minutes to roughly one week. Approval for chronic weight management followed in 2021, based on large cardiovascular and obesity outcome trials.

=== The Americas === Europeans introduced saffron to the Americas when immigrant members of the Schwenkfelder Church left Europe with a trunk containing its corms. Church members had grown it widely in Europe. By 1730, the Pennsylvania Dutch cultivated saffron throughout eastern Pennsylvania. Spanish colonies in the Caribbean bought large amounts of this new American saffron, and high demand ensured that saffron's list price on the Philadelphia commodities exchange was equal to gold. Trade with the Caribbean later collapsed in the aftermath of the War of 1812, when many saffron-bearing merchant vessels were destroyed. Yet the Pennsylvania Dutch continued to grow lesser amounts of saffron for local trade and use in their cakes, noodles, and chicken or trout dishes. American saffron cultivation survives into modern times, mainly in Lancaster County, Pennsylvania.
Birds have featured in culture and art since prehistoric times, when they were represented in early cave painting and carvings. Some birds have been perceived as monsters, including the mythological Roc and the Māori's legendary Pouākai, a giant bird capable of snatching humans. Birds were later used as symbols of power, as in the magnificent Peacock Throne of the Mughal and Persian emperors. With the advent of scientific interest in birds, many paintings of birds were commissioned for books. Among the most famous of these bird artists was John James Audubon, whose paintings of North American birds were a great commercial success in Europe and who later lent his name to the National Audubon Society. Birds are also important figures in poetry; for example, Homer incorporated nightingales into his Odyssey, and Catullus used a sparrow as an erotic symbol in his Catullus 2. The relationship between an albatross and a sailor is the central theme of Samuel Taylor Coleridge's The Rime of the Ancient Mariner, which led to the use of the term as a metaphor for a 'burden'. Other English metaphors derive from birds; vulture funds and vulture investors, for instance, take their name from the scavenging vulture. Aircraft, particularly military aircraft, are frequently named after birds. The predatory nature of raptors make them popular choices for fighter aircraft such as the F-16 Fighting Falcon and the Harrier Jump Jet, while the names of seabirds may be chosen for aircraft primarily used by naval forces such as the HU-16 Albatross and the V-22 Osprey.
Flavoproteins are proteins that contain a nucleic acid derivative of riboflavin. These proteins are involved in a wide array of biological processes, including removal of radicals contributing to oxidative stress, photosynthesis, and DNA repair. The flavoproteins are some of the most-studied families of enzymes. Flavoproteins have either FMN (flavin mononucleotide) or FAD (flavin adenine dinucleotide) as a prosthetic group or as a cofactor. The flavin is generally tightly bound (as in adrenodoxin reductase, wherein the FAD is buried deeply). About 5-10% of flavoproteins have a covalently linked FAD. Based on the available structural data, FAD-binding sites can be divided into more than 200 different types. 90 flavoproteins are encoded in the human genome; about 84% require FAD and around 16% require FMN, whereas 5 proteins require both. Flavoproteins are mainly located in the mitochondria. Of all flavoproteins, 90% perform redox reactions and the other 10% are transferases, lyases, isomerases, ligases.
=== Social media and memes === On X (formerly Twitter), there are numerous accounts with names like "Socialists for Pritzker", "Nomadic Warriors for Pritzker", "Frat Bros for Pritzker", and "Anarchists for Pritzker". Socialists for Pritzker has over 13,000 followers. The accounts have been described as semi-ironic. Pritzker has said the attention is "entertaining, if a little strange" and that he keeps up with several of the accounts. Nomadic Warriors for Pritzker has dubbed Pritzker the "Great Khan of the Midwest" and mapped out battle plans for a Mongol Empire-inspired military campaign across the nation. The group of accounts, dubbed the "Pritzker Pals", launched a website, coconutbigboy.com, with the goal of influencing Vice President Kamala Harris to pick Pritzker as her running mate in the 2024 presidential election. A TikTok account called "Pritzker Memes" garnered 20,000 followers. Some of the memes relate to Pritzker's size, with one political commentator saying, "He is enormous, doesn't come off as particularly intellectual, and has good instincts". Pritzker launched his own beer brand, JBeers, ahead of the 2024 Democratic National Convention in Chicago. He also named Malört the DNC's "unofficial shot". On February 7, 2025, after President Trump announced that the Gulf of Mexico had been renamed the "Gulf of America", Pritzker posted a satirical video on his personal Twitter account announcing Illinois's annexation of Green Bay, Wisconsin, and the renaming of Lake Michigan to "Lake Illinois".

=== Post-settlement activity === In 1991, the local Bhopal authorities charged Anderson, who had retired in 1986, with manslaughter, a crime that carries a maximum penalty of 10 years in prison. He was declared a fugitive from justice by the Chief Judicial Magistrate of Bhopal on 1 February 1992 for failing to appear at the court hearings in a culpable homicide case in which he was named the chief defendant. Orders were passed to the Government of India to press for an extradition from the United States. The United States declined to extradite him citing a lack of evidence. The U.S. Supreme Court refused to hear an appeal of the decision of the lower federal courts in October 1993 against allowing victims of the Bhopal disaster to seek damages in a U.S. court. In 2004, the Indian Supreme Court ordered the Indian government to release any remaining settlement funds to victims. And in September 2006, the Welfare Commission for Bhopal Gas Victims announced that all original compensation claims and revised petitions had been "cleared". In 2006, the U.S. Court of Appeals for the Second Circuit in New York City upheld the dismissal of remaining claims in the case of Bano v. Union Carbide Corporation. This move blocked plaintiffs' motions for class certification and claims for property damages and remediation.
== Diseases == The inappropriate activation of trypsinogen in the pancreas can lead to pancreatitis. Some type of pancreatitis may be associated with mutant forms of trypsinogen. A mutation at Arg 117, a trypsin-sensitive site, in cationic trypsinogen has been implicated in hereditary pancreatitis, a rare form of early-onset genetic disorder. Arg 117 may be a fail-safe mechanism by which trypsin, when activated within the pancreas, may become inactivated, and loss of this cleavage site would result in a loss of control and permit autodigestion resulting in pancreatitis. Other mutations have also been found that are linked to pancreatitis.
The planters of the Dutch colony of Suriname relied heavily on African slaves to cultivate, harvest and process the commodity crops of coffee, cocoa, sugar cane and cotton plantations. The Netherlands abolished slavery in Suriname in 1863. Many slaves escaped the plantations. With the help of the native South Americans living in the adjoining rain forests, these runaway slaves established a new and unique culture in the interior that was highly successful in its own right. They were known collectively in English as Maroons, in French as Nèg'Marrons (literally meaning "brown negroes", that is "pale-skinned negroes"), and in Dutch as Marrons. The Maroons gradually developed several independent tribes through a process of ethnogenesis, as they were made up of slaves from different African ethnicities. These tribes include the Saramaka, Paramaka, Ndyuka or Aukan, Kwinti, Aluku or Boni, and Matawai. The Maroons often raided plantations to recruit new members from the slaves and capture women, as well as to acquire weapons, food and supplies. They sometimes killed planters and their families in the raids. The colonists also mounted armed campaigns against the Maroons, who generally escaped through the rain forest, which they knew much better than did the colonists. To end hostilities, in the 18th century the European colonial authorities signed several peace treaties with different tribes. They granted the Maroons sovereign status and trade rights in their inland territories, giving them autonomy.

Long-term storage is usually at minus 20 to minus 80 degrees Celsius in a sealed, desiccated container. Working aliquots can be held briefly at 2 to 8 degrees Celsius.
Repeated temperature cycling can promote aggregation and peptide degradation. Dividing material into single-use aliquots limits this risk.
Mass spectrometry is commonly used to confirm molecular mass and detect structural modifications. It is often paired with chromatographic purity assessment.
It is given either as a once-weekly subcutaneous injection or as an oral tablet taken once daily. The two forms use different absorption strategies, so they are not interchangeable on a milligram-for-milligram basis.
