Handling practices center on minimizing contamination and adsorption. Lyophilized peptide tends to accumulate static charge, so weighing is done with antistatic measures and calibrated balances. Reconstitution with appropriate solvent should be gentle, avoiding vigorous vortexing that generates foam and shear. Solutions are typically aliquoted before freezing to reduce repeated temperature cycling. Personal protective equipment and a fume hood are standard for powder handling.
Reconstituted solutions are less stable than the dry powder, and stability depends on concentration, pH, buffer composition, and container material. Low-protein-binding tubes reduce loss of peptide to plastic surfaces. Some researchers add a carrier protein to limit adsorption at low concentrations. The exact shelf life of a given solution is best determined empirically through a stability study rather than assumed from general guidance, because published data cover only a limited set of conditions.
Storage conditions for semaglutide depend heavily on the presentation. Lyophilized research powder is generally kept at two to eight degrees Celsius in a sealed container, protected from light and moisture. Manufacturer labeling for finished injectable products specifies refrigeration before first use, with defined in-use periods at room temperature afterward. The oral tablet form is stored at controlled room temperature and is more tolerant of short excursions. Temperature excursions should be documented rather than inferred.
Clinical studies of semaglutide generally measure glycated hemoglobin, fasting plasma glucose, body weight, and composite cardiovascular endpoints. The SUSTAIN program enrolled adults with type 2 diabetes, while the STEP program focused on obesity without diabetes. Administration follows a stepwise escalation schedule designed to limit gastrointestinal effects during the first weeks. Reported outcomes include mean percentage weight change, the proportion of participants reaching defined weight-loss thresholds, and rates of nausea, vomiting, and diarrhea. Long-term data on durability after treatment stops are still limited and remain a topic of ongoing research.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, developed by Novo Nordisk and first approved in 2017 for type 2 diabetes. It belongs to the incretin mimetic class, a group of agents that reproduce the glucose-dependent actions of endogenous GLP-1. The molecule was engineered to resist degradation by dipeptidyl peptidase-4 and to bind serum albumin, extending its half-life from minutes to roughly one week. Approval for chronic weight management followed in 2021, based on large cardiovascular and obesity outcome trials.

Reverse-phase high-performance liquid chromatography with ultraviolet detection near 214 nm is the standard purity method, reported as area percent. Mass spectrometry, usually with electrospray ionisation, confirms identity and reveals covalent modifications. Size-exclusion chromatography quantifies aggregates and fragments. Peptide mapping after enzymatic digestion localises changes to specific sequence regions. Circular dichroism and infrared spectroscopy report on secondary structure, while light scattering tracks particle formation in liquid formulations. No single technique captures every quality attribute.
Quality control relies on pharmacopoeial monographs where they exist, combined with in-house specifications for identity, purity, water content, and counter-ion composition. Reference standards allow calibration across laboratories, although certified materials for every analogue are not universally obtainable. Batch records, chromatograms, and mass spectra form the documentation trail. Regulatory classification varies by jurisdiction and intended use, and research-grade material differs from pharmaceutical-grade material in testing scope. Analytical uncertainty is often expressed as relative standard deviation across replicate injections.
Handling guidance for research quantities calls for single-use aliquots, an inert atmosphere where practical, and avoidance of repeated freeze-thaw cycles that accelerate aggregation. Certificates of analysis typically report purity by peak area, water content, counter-ion identity, and residual solvent levels. In the scientific literature the compound is usually described by its full amino acid sequence, its registry number, or its structural class rather than by any proprietary label. Reporting standards vary between journals, and reviewers increasingly request raw chromatograms alongside tabulated purity figures. Whether current purity thresholds are adequate for every experimental context is debated.
Reversed-phase high-performance liquid chromatography with ultraviolet detection is the dominant approach for peptide purity assessment, usually paired with mass spectrometry to confirm molecular mass and sequence. Peptide mapping by enzymatic digestion and tandem mass spectrometry locates modifications such as deamidation and oxidation. Quantitation in plasma matrices can be performed by LC-MS/MS after solid-phase extraction. Method validation follows general guidance on accuracy, precision, linearity, and limits of detection. Comparability of results between laboratories, when no shared reference standard is available, remains an open question.
Development began in the early 2010s with the goal of extending GLP-1 activity beyond the brief window achieved by native peptide infusion. The earliest approved formulation was a subcutaneous injection given once weekly. A later oral tablet pairs the peptide with an absorption enhancer, sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, usually shortened to SNAC. That carrier lowers local pH and helps the peptide cross gastric tissue. Both routes deliver the same active molecule.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, a gut hormone released after meals. Its backbone retains the GLP-1 sequence but incorporates two substitutions that slow enzymatic breakdown by dipeptidyl peptidase-4. A short polyethylene glycol linker and a C18 fatty diacid are attached to the peptide chain, allowing the molecule to bind serum albumin and remain in circulation far longer than the native hormone. The result is a circulating half-life measured in days rather than the minutes typical of endogenous GLP-1.
Δh = The head loss due to pipe friction over the given length of pipe (SI units: m); g = The local acceleration due to gravity (m/s2). It is useful to present head loss per length of pipe (dimensionless):
Before the 1967 war, Christians comprised 12% of the total population of the Golan, which reached 150,000 people. While Druze formed the majority of the population. Christians were involved in small-scale economic and commercial activities, classified within the petite bourgeoisie, and most were educated, with many working in professions such as medicine, law, and engineering, while the Druze mainly worked in agriculture and on the land. The vast majority of Christians migrated with the rest of the population after Israel's occupation of the Golan, leaving only a few small Christian families. Of the four remaining Syrian Druze communities in the Israeli-occupied territories (on Israel's side of Mount Hermon and the Golan Heights), Majdal Shams is the largest, together with Ein Qiniyye, Mas'ade, and Buq'ata. As of 2017, there was one Greek Orthodox Christian family of five (the Nasrallah family) in Majdal Shams, and one Christian family of twelve (the Assaf family) in Ein Qiniyye. Only one Maronite church remains in Ein Qiniyye, along with remnants of two Melkite churches in Ein Qiniyye and one Orthodox church in Majdal Shams. The historic church of Banias remains closed despite restoration efforts. Relations between Christians and Druze in Majdal Shams and Ein Qiniyye are characterized by goodwill, peaceful coexistence, and mixing.
There appears to be no consensus on the existence of substances that may have at one time been named as vitamins Q, R, T, V, W, X, Y or Z. "Vitamin N" is a term popularized for the mental health benefits of spending time in nature settings. "Vitamin I" is slang among athletes for frequent/daily consumption of ibuprofen as a pain-relieving treatment.
== Personal life == Austin played college football at the University of North Texas. Austin married his high school girlfriend Kathryn Burrhus on November 24, 1990. However, he later pursued a relationship with English wrestling manager Jeanie Clarke, with whom he was working. His marriage to Burrhus was annulled on August 7, 1992, while he was in Japan and he married Clarke on December 18, on his 28th birthday. They had two daughters before divorcing in 1999. Austin also adopted Clarke's daughter from a previous relationship with Chris Adams. Austin's adoptive daughter was married to fellow professional wrestler Adam Bryniarski until his death in 2022. In 1998, Austin was the recipient of Hamilton, Ontario's "Key to the City", he is one of two people to ever be bestowed the honor, the other being the basketball player Shai Gilgeous-Alexander. That same year on September 1, the city of Lowell, Massachusetts declared that date Stone Cold day along with naming a street after him for the day. The date March 16 is commonly associated with Austin due to the date matching up with his wrestling catchphrase "Austin 3:16". On September 13, 2000, Austin married wrestling manager Debra Marshall. On June 15, 2002, Marshall called the police to the couple's home. She told officers that Austin had hit her and then stormed out of the house before police arrived. An arrest warrant was issued by the Bexar County district attorney's office on August 12 and Austin turned himself in the following day, at which point he was charged with domestic abuse.
== History == DET was first synthesized and administered intramuscularly in a 60 mg dose by Stephen Szára in 1956. It was subsequently described in his material published in 1957. More systematic studies were reported later by Szara and colleagues and independently by Böszörményi and colleagues. Early research began as a search for "psychosis mimics" in psychiatry, then expanded into broader psychedelic and structure–activity studies. Selection of study subjects for some of these studies was criticized by Alexander Shulgin in his 1997 book TiHKAL (Tryptamines I Have Known and Loved) for its "oppressive research environment". For many years, based on early clinical reports and private communications, Shulgin maintained that DET exhibited psychoactive effects only when administered via parenteral routes. He eventually revised his view, ultimately acknowledging that the substance is also orally active. Initially, DET was not classified as a controlled substance, and some early clinical and experimental psychopharmacological research used it without scheduling restrictions. By the late 1960s and early 1970s, however, increasing regulatory attention led to tighter controls and this led to DET getting placed in Schedule I internationally by the Convention on Psychotropic Substances. Modern research on DET remains limited compared to dimethyltryptamine (DMT), due to its status as a controlled substance and the predominance of focus on other tryptamines with greater prevalence in traditional or clinical contexts.

Calendula () is a genus of about 15–20 species of annual and perennial herbaceous plants in the daisy family, Asteraceae that are often known as marigolds. They are native to Europe, North Africa, Macaronesia, West Asia and Western Himalayas and have their center of diversity in the Mediterranean Region. Other plants known as marigolds include corn marigold, desert marigold, marsh marigold, and plants of the genus Tagetes. The genus name Calendula is a modern Latin diminutive of calendae, meaning "little calendar", "little clock" or possibly "little weather-glass". The common name "marigold", a contraction of "Mary's gold" used especially for Calendula officinalis, refers to the Virgin Mary. C. officinalis is the most commonly cultivated and used species, popular herbal and cosmetic products named "Calendula" invariably derive from it.
==== NADP ==== Nicotinamide adenine dinucleotide phosphate is a coenzyme present in redox and biosynthetic reactions. The domain binds NADP in its oxidised or reduced forms as NADP+ or NADPH respectively.
== DeSUMOylation == SUMO can be removed from its substrate, which is called deSUMOylation. Specific proteases mediate this procedure (SENP in human or Ulp1 and Ulp2 in yeast). In yeast, SMT3 encodes the SUMO protein, and SUMO E3 ligase attaches SUMO to target proteins. In cell cycle regulation, the base case is that SUMO ligation is constantly taking place, leading to polySUMOylation of eligible target proteins. This is countered by the SUMO protease Ulp2 which cleaves polySUMO groups, leaving the protein in a monoSUMOylated state. As shown in the figure, there is a feedback mechanism in which ULP2 maintains the monoSUMOylated state by passively and diligently cleaving SUMO such that the polySUMOylated state is never stabilized enough to be acted upon by downstream actors. This deSUMOylation is critical to prevent precocious advancement of the cell cycle as discussed in several studies.
==== Immunological biosensor ==== Gold nanoparticles have been coated with peptides and glycans for use in immunological detection methods. The possibility to use glyconanoparticles in ELISA was unexpected, but the method seems to have a high sensitivity and thus offers potential for development of specific assays for diagnostic identification of antibodies in patient sera.

Hydrogen selenide is an extremely toxic, corrosive gas. Selenium also occurs in organic compounds, such as dimethyl selenide, selenomethionine, selenocysteine and methylselenocysteine, all of which have high bioavailability and are toxic in large doses. The Occupational Safety and Health Administration (OSHA) has set the legal limit (permissible exposure limit) for selenium in the workplace at 0.2 mg/m3 over an 8-hour workday. The National Institute for Occupational Safety and Health (NIOSH) has set a Recommended exposure limit (REL) of 0.2 mg/m3 over an 8-hour workday. At levels of 1 mg/m3, selenium is immediately dangerous to life and health.
Acid proteases Neutral proteases involved in type 1 hypersensitivity. Here, it is released by mast cells and causes activation of complement and kinins. This group includes the calpains. Basic proteases (or alkaline proteases)
In July 2016, a leak of the Democratic National Committee's emails appeared to show DNC officials favoring Clinton over Sanders. Staff repeatedly discussed making his irreligious tendencies a potential campaign issue in southern states and questioned his party loyalty. DNC chair Debbie Wasserman Schultz called his campaign manager "an ass" and "a damn liar". Speaking with Jake Tapper on CNN, Sanders responded to the leak, saying, "it is an outrage and sad that you would have people in important positions in the DNC trying to undermine my campaign. It goes without saying: the function of the DNC is to represent all of the candidates—to be fair and even-minded. But again, we discussed this many, many months ago, on this show, so what is revealed now is not a shock to me."
== Research in cancer == Deoxycholate and other secondary bile acids cause DNA damage. Secondary bile acids increase intracellular production of reactive oxygen and reactive nitrogen species resulting in increased oxidative stress and DNA damage. As shown in the figure below, deoxycholate added to the diet of mice increased the level of 8-oxo-dG, an oxidative DNA damage, in the colonic epithelium of mice. When the level of deoxycholate-induced DNA damage is high, DNA repair enzymes that ordinarily reverse DNA damage may not be able to keep up. DNA damage has frequently been proposed as a major cause of cancer. DNA damage can give rise to cancer by causing mutations. When deoxycholate was added to the food of mice so that their feces contained deoxycholate at about the same level present in feces of human on a high fat diet, 45% to 56% of the mice developed colon cancer over the next 10 months, while none of the mice on a diet without deoxycholate developed cancer. Thus, exposure of the colon to deoxycholate may cause cancer in mice. However, this same study reported that, when chlorogenic acid was added to the diet alongside deoxycholate, only 18% of the mice developed colon cancer. Chlorogenic acid is a component of common foods and beverages; coffee contains an average of 53.8 mg chlorogenic acid per 100 mL. Therefore, to consume the level of chlorogenic acid used in the study, a human on a "standard" 2000-calorie daily diet (416 g/d; 250 g carbs, 100 g protein, 66 g fat) would need to consume roughly 55 mL of coffee each day, or just under 2 fluid ounces.
Values for other compounds may be found in a variety of available reviews and monographs. Critical discussions of the challenges of measurement of log P and related computation of its estimated values (see below) appear in several reviews.

Sealed, protected from light, and refrigerated at two to eight degrees Celsius for most research material. Desiccated storage limits moisture uptake. Allow the vial to reach room temperature before opening to prevent condensation.
Each cycle exposes the peptide to ice-liquid interfaces where unfolding and aggregation can occur. Aggregates may not redissolve and can alter measured activity. Aliquoting before the first freeze reduces the number of cycles any single portion experiences.
It usually reports purity by HPLC, identity by mass spectrometry, peptide content, appearance, and residual moisture or counterion content. The analytical method behind each value should be stated. Acceptance criteria are set by the supplier or the buyer's specification.
It is given either as a once-weekly subcutaneous injection or as an oral tablet taken once daily. The two forms use different absorption strategies, so they are not interchangeable on a milligram-for-milligram basis.
